Pedagogical objectives

Recent and ongoing advances in optical imaging are opening new avenues for studying intact biological tissues with subcellular resolution. These advances are based on sophisticated physical concepts and technologies aiming at overcoming current limitations in imaging depth, speed, volume, specificity, etc…. They provide unique information in experimental system biology, mainly in developmental biology, neurosciences, as well as in physiopathology.

This course will focus both on the physics of these advanced microscopies and on specific applications exemplifying new biological and medical knowledges provided by these techniques.

 

Program

This course builds on the “Biophotonics and Applications” course BIO_52004_EP and expands to more complex techniques and applications.

  • Light propagation and image formation - Holography
  • Focusing theory: analytical and numerical methods
  • Aberrations and wavefront shaping – Application to retina imaging
  • Advanced multiphoton microscopy (1): coherent and polarization-resolved signals - Applications to connective tissues (skin, cornea)
  • Advanced multiphoton microscopy (2): large volume and high depth techniques - Applications to neurosciences
  • Imaging of awake small animals and fibroscopy – Application to neurosciences
  • Fast and high-throughput optical imaging; Light-sheet microscopy – Applications to developmental biology
  • Advanced concepts and methods for super-resolution – Applications to cellular imaging.

 

Pre-requisites

BIO_52004_EP - Biophotonics and Applications must be taken in parallel.
